چکیده

The scientific literature contains an ever-growing number of reports of applications of vibrational spectroscopy as a multivariate non-invasive tool for analysis of biological effects at the molecular level. Recently, Fourier transform infrared microspectroscopy (FTIRM) has been demonstrated to be sensitive to molecular events occurring in cells and tissue after exposure to ionizing radiation. In this work the application of FTIRM in the examination of dose-dependent molecular effects occurring in skin cells after exposure to ionizing radiation with the use of partial least-squares regression (PLSR) and generalized regression neural networks (GRNN) was studied. The methodology is shown to be sensitive to molecular events occurring with radiation dose and time after exposure. The variation in molecular species with dose and time after irradiation is shown to be non-linear by virtue of the higher modeling efficiency yielded from the non-linear algorithms. Dose prediction efficiencies of approximately +/-10 mGy were achieved at 96 h after irradiation, highlighting the potential applications of the methodology in radiobiological dosimetry.

Fourier Transform Infrared (FTIR) Microspectroscopy of Immature, Mature and Degenerated Articular Cartilage

Osteoarthritis is a common joint disease characterized by the progressive degeneration of the articular cartilage. In this chapter, the application of Fourier transform infrared (FTIR) microspectroscopy for the determination of spatial composition of main structural components of articular cartilage, i.e. collagen and proteoglycans, is presented. The previous results from FTIR microspectroscopi...

The Characterisation of Pluripotent and Multipotent Stem Cells Using Fourier Transform Infrared Microspectroscopy

Fourier transform infrared (FTIR) microspectroscopy shows potential as a benign, objective and rapid tool to screen pluripotent and multipotent stem cells for clinical use. It offers a new experimental approach that provides a holistic measurement of macromolecular composition such that a signature representing the internal cellular phenotype is obtained. The use of this technique therefore con...

Rapid analysis of two food-borne microbial communities at the species level by Fourier-transform infrared microspectroscopy.

The species composition of microbial communities in natural habitats may be extremely complex and therefore a quantitative analysis of the fraction each species contributes to the consortium has proven to be difficult. During recent years, the identification of bacterial pure cultures based on their infrared spectra has been established. Fourier-transform infrared microspectroscopy now proceeds...

FTIR Microspectroscopy of Polymeric Systems

Fourier transform infrared (FTIR) spectroscopy is a mature analytical technique employed to examine polymeric materials. With the coupling of an infrared interferometer to a microscope equipped with specialized detectors, FTIR spectroscopy has been employed widely to examine microscopic areas in polymers for the last twenty years. Following the emergence of instrumentation techniques that emplo...

Non-Invasive Fourier Transform Infrared Microspectroscopy and Imaging Techniques: Basic Principles and Applications

The current paper describes rather new and straightforward non-invasive mid infrared spectroscopic techniques for the position sensitive identification of sample components. These techniques include mid infrared (MIR) microspectroscopy, mapping as well as imaging approaches e.g. in transmission, reflection-absorption, or attenuated total reflection mode. The basic principles of the different in...
